Streptococcus is a spherical Gram-positive bacteria. It is characterized via Lancefield serotyping, which describes specific carbohydrates present on the bacterial cell wall. 20 serotypes have been described and named Lancefield groups A to V (excluding I and J). The ImmuLex™ Streptococcus Group Kit is a fast latex agglutination test for serotyping streptococci within groups A, B, C, D, F and G, which are most prevalent.
ImmuLex™ Streptococcus Group Kit is used for serotyping of Streptococcus(groups A, B, C, D, F and G) by latex agglutination. The group kit consists of six vials each containing 1.5 mL latex particles coated with streptococcal group antiserum produced in rabbits (group A, B, C, D, F and G). Cross-reactions in the antisera have been removed by absorption. Furthermore, it contains three extraction reagents: extraction reagent 1, containing two vials of 2 mL sodium nitrite solution, extraction reagent 2, containing two vials of 2 mL acid solution, and extraction reagent 3, containing two vials of 2 mL of neutralizing solution. Reaction cards and mixing sticks for latex agglutination are also included in the group kit. The group kit contains material sufficient for 75 tests.Antisera and extraction reagents should be stored at 2–8 °C degrees. The six group antisera and extraction reagents can also be purchased separately.
The ImmuLex™ Streptococcus Group Kit is a rapid latex agglutination test for streptococcal serotyping using chemical extraction of group specific antigens with nitrous acid extraction reagents. Both the extraction and serotyping is performed at room temperature.
An agglutination test is performed by mixing group antiserum and latex reagent on a reaction card according to instructions. If there is no reaction or if reaction is indistinct the bacteria is extracted with the three reagents according to instructions. A positive reaction will be visible within 30 seconds. If there is no positive reaction within 30 seconds the test is negative. A positive reaction in one of the latex reagents identifies the Streptococcus group. If there is only a positive reaction in group B reagent, the bacteria is a Streptococcus group B strain.
